What Is Argireline

Argireline was developed as a breakthrough topical alternative to botulinum toxin. However, unlike botulinum toxin (“Botox in a bottle”), which requires subcutaneous injection, Argireline works by topical application to the epidermis, thereby avoiding the risks associated with subcutaneous injections.

At its core, Argireline interferes with the neurotransmitter release by targeting the SNARE complex (soluble NSF attachment protein receptor),which is essential for docking acetylcholine containing vesicles.Through this mechanism, it disrupts the SNARE complex’s formation and stability, thereby inhibiting neurotransmitter release.

Building on this mechanism, notably, scientists modeled Argireline on the N-terminus structure of the SNAP-25 protein . It competes with SNAP-25 for binding to vesicle-associated membrane protein (VAMP/synaptobrevin).This peptide technology disrupts the formation of the SNARE complex, thereby inhibiting neuronal exocytosis  [1].

1.Reduction of Facial Muscle Contraction

Argireline minimizes the appearance of expression lines(like forehead wrinkles and crow’s feet) by inhibiting acetylcholine release during exocytosis, which prevents muscle contraction. For example,clinical Studies have shown that 10% argireline serum can reduce wrinkle depth[2]  by 30% after 30 days in vivo.

2.Improvement in Skin Texture and Smoothness

Clinical studies have shown that applying a formulation containing 10% acetyl hexapeptide-8 to facial skin daily for 20 days can slightly reduce skin roughness and minimizes transepidermal water loss (TEWL) [3]  .Furthermore,after 4 weeks, the formulation reduced wrinkle depth by 49% [4] .

Most notably, before and after treatment photos showed significant improvement in scars caused by surgical interventions related to cancer treatment, cyst removal, or mole removal.
