What is changing in pharmaceutical manufacturing, and why does it reach peptide suppliers?

Annex VII of the Industrial Emissions Directive covers solvent-using activities, including pharmaceutical manufacturing above its specified consumption threshold. Applicable emission limits, exclusions, national permits and the solvent-management plan must be read together; the program itself does not impose a solvent ban. Read the Directive.

The commercial driver comes from your own customers. EFPIA’s survey of 23 member companies found that 95% engage with suppliers on climate action, more than 70% hold long-term Scope 3 reduction targets, and about 45% have specified measurable short-term Scope 3 targets (EFPIA’s survey of 23 member companies).
A supplier’s own solvent consumption and reporting obligations are now part of the buyer’s Scope 3 arithmetic, which is why a programme like ETERNAL reaches a peptide vendor that never signed up to it.
Which peptide-synthesis solvents are under pressure, and what are the validated alternatives?

There is no single universal solvent that replaces DMF. The ACS GCI Pharmaceutical Roundtable’s greener peptide synthesis guide states that viable substitution is often a mixture, that the optimum depends on each resin and peptide sequence, and that candidates must be screened case by case; documented failure modes include undesirable resin swelling, bed-volume changes, insolubility of amino-acid synthons, and lower yield and purity (the Roundtable’s own list of DMF replacements). The same guide names Cyrene™, sulfolane or anisole with dimethyl or diethyl carbonate, NBP (non-CMR, but too costly for large-scale manufacture and too viscous neat), NOP at roughly $1 to $2/kg, NOP/dimethyl carbonate, NBP/2-MeTHF, NBP/1,3-dioxolane, N-formyl morpholine/DOL, and DMSO/EtOAc mixtures.
द 2026 review of DMF-free routes adds the performance caveats that matter at the bench: DMSO performs in some cases approaching DMF, improves coupling with sterically hindered amino acids and reduces racemisation, but has low swelling capacity and reduces reaction rates and resin accessibility, while 2-MeTHF and EtOAc appear only as components of binary mixtures rather than standalone replacements (द 2026 review of DMF-free routes).

The mechanism behind several of these options is resin rigidity. The multifactorial greening study, which found non-swelling resins cut solvent consumption by at least half, explains why: a single synthetic cycle may require at least ten washing steps, each using roughly Dipeptide Manufacturer ten volumes of solvent relative to the solid support, and a support that does not swell removes the constraint that dictated DMF in the first place (the multifactorial greening study).
Screen candidates on the actual sequence, and treat any claimed drop-in replacement as unvalidated until it is.
How do you validate a solvent swap without losing purity, impurity control or yield?
A change to a validated analytical procedure triggers a revalidation question, not an automatic full re-run. I Q2(R2), adopted in November 2023, states in Section 2.2 that partial or full revalidation may be required, that science- and risk-based principles can justify whether a given performance characteristic needs revalidation, and that the extent depends on the characteristics impacted by the change (I Q2(R2), adopted in November 2023). ICH Q14, adopted the same day, describes the science- and risk-based development approach that supports that justification (ICH Q14).
The chemistry side is where swaps actually fail. The wash-free SPPS work found pyrrolidine at 3 को 3.5% optimal, with crude purity of 86% र 84% under those conditions, and reported that piperidine under identical wash-free conditions gave 75% against 84% for pyrrolidine because of lower reactivity and evaporation rate (the wash-free SPPS work). A vendor application note on the ACP65-74 10mer shows the same interaction from the other direction: cutting to 1 post-deprotection wash and 2 needle washes gave 76.6% crude purity with 20% piperidine/DMF, र 9.5% when the base was switched to 2% DBU/5% piperazine, recovering only to 62.3% with a 1% Oxyma additive (a vendor application note on the ACP65-74 10mer). The base and the wash strategy cannot be changed independently.
टिप को लागी: Compare the same sequence, same resin loading, same coupling time and same analytical method. Report crude purity, impurity profile and isolated yield against the incumbent, and record explicitly whether aggregation, incomplete deprotection, epimerisation or purification difficulty changed.
A solvent swap is accepted only when the analytical package, not the solvent label, shows equivalence.
Where does the environmental burden really sit — synthesis, शुद्धीकरण, or lyophilization?
The ACS GCI Pharmaceutical Roundtable’s PMI analysis of 40 peptide processes, drawn from 14 member companies, reports an average SPPS process mass intensity of 13,063 kg/kg across 14 commercial and Phase 3 पेप्टाइड्स, with a range of 1,684 को 34,585 for peptides of 6 को 43 एमिनो एसिड (the ACS GCI Pharmaceutical Roundtable’s PMI analysis of 40 peptide processes). Process mass intensity is kilograms of input per kilogram of product. For scale, small molecules show a PMI median of 168 को 308 and biopharmaceuticals roughly 8,300 on the same basis.
The counter-intuitive finding is where that mass goes. Downstream processes account directly for approximately 50% of PMI, because purification uses dilute product solutions and long chromatographic gradients, and purification yield feeds back into overall PMI since downstream losses must be offset upstream. Solvents and water are used in approximately equal amounts, 44% each, and DMF is the dominant organic solvent at 39% of process waste.
That reframes the wash-reduction argument. Post-deprotection washings account for approximately 90% of the waste generated in SPPS, and a 25 mmol wash-free liraglutide synthesis generated 28.4 L of total waste against 139.7 L for the wash-based run, an overall reduction of approximately 80% (the wash-free SPPS work that cut a 25 mmol liraglutide run from 139.7 L of waste to 28.4 एल). But any crude-purity loss from fewer washes is paid for twice: once in yield, and again in the extra chromatography needed to reach specification. The Biotage figure of over 50% total system solvent reduction is the most conservative and most explicitly bounded of the published claims, because it states what is and is not counted.
Do not split that 50% downstream share into HPLC versus lyophilization components; the source does not, and lyophilization is efficient from a PMI perspective as a standalone unit operation even though it is energy intensive.
Which metrics should a peptide buyer ask for, and what do they mean per batch?
Ask for four metrics, defined before you accept any number. Process mass intensity is total mass input divided by mass of product, expressed in kg/kg. Solvent intensity is the solvent-only subset of that input. E-factor is mass of waste divided by mass of product, which is the correct metric for a waste-per-kilogram claim and is not interchangeable with PMI. Solvent recovery rate is recovered solvent returned to the process as a share of solvent input, and it maps directly onto the I2 and O8 categories of a solvent management plan.
The reason to insist on definitions is that published figures are not comparable across bases. The ultrasound-assisted SPPS paper reports 83 को 88% solvent reduction per coupling cycle and an 88.6% cost saving at September 2024 DMF prices, with PMI and E-factor of 385.29 र 369.56 against 2,915.90 र 2,900.16 for standard SPPS on a 0.5 mmol pentapeptide (ultrasound-assisted SPPS, which cut solvent use per coupling cycle by 83 को 88%). Those are single-molecule, sub-millimolar figures. The PMI study covers commercial and Phase 3 processes up to 34,585 kg/kg. Neither number transfers to the other’s case.

What sustainability data can a supplier actually disclose, and how do you request it?
For an installation within scope, use the applicable solvent-management plan and permit requirements as one evidence source. Annex VII distinguishes inputs I1 and I2 and outputs O1 through O9. O8 covers recovered solvent for reuse outside the input category, and O9 other releases. A site-period balance is not automatically a batch-level peptide balance.
Request a documented mass balance with a defined period, process boundary and product denominator. Purchased solvent and collected waste alone cannot determine total solvent intensity: account for inventory changes, recovered inputs, allocation and product output as appropriate.
Two documentation rules from the customer’s own quality framework apply directly here. A purity percentage alone does not establish complete compound identity; शुद्धता, पहिचान, विधि, lot number and documentation should be evaluated together. And a peptide certificate of analysis should be reviewed as a batch-specific record, not a general marketing statement. Apply both to any sustainability figure you receive: a programme-level percentage is a marketing statement, a batch-level mass balance is a record.
Where the evidence stops, say so. No supplier sustainability questionnaire standard was identified in the sources reviewed, so the Annex VII categories are a template, not a certification scheme.
How do you keep purity, पहिचान, content and lot-to-lot consistency requirements intact while asking for greener routes?
Separate the specification from the route. शुद्धता, पहिचान, content and lot-to-lot consistency are release criteria; the solvent system is a process parameter. A greener route is acceptable only when the release criteria are met on the same analytical package, and the revalidation logic in ICH Q2(R2) tells you how to scope that: identify which performance characteristics the change actually impacts, and justify the extent of revalidation on that basis rather than re-running everything (I Q2(R2), adopted in November 2023).
Set acceptance criteria before the comparison, including relevant stereoisomers and process impurities. Per-residue stereochemical fidelity does not alone establish the total epimer level or compliance with a particular impurity limit; measure the relevant impurities in the product.
Then hold the route constant across lots. A solvent swap validated on one lot and not re-checked on the next is a process change without a change control. The decision rule is simple: a greener route is accepted when the batch record shows the same release criteria met by the same methods, and rejected when the analytical package is thinner than the incumbent’s.

Does ETERNAL mean peptide suppliers will be required to change solvents?
छैन. ETERNAL is a research and innovation programme, and the figures attributed to it are reported expected results from pilots, not requirements. The binding instrument is Annex VII of the Industrial Emissions Directive, which applies to pharmaceutical manufacturing above 50 tonnes per year of solvent consumption and requires a solvent management plan rather than a specific solvent choice (Annex VII of the Industrial Emissions Directive).
Can DMF be replaced with a single greener solvent?
Not according to the ACS GCI Pharmaceutical Roundtable, which states there is no single universal replacement and that viable substitution is often a mixture screened case by case against the resin and sequence (the Roundtable’s own list of DMF replacements). DMSO comes closest on coupling performance but has low swelling capacity and reduces reaction rates and resin accessibility (द 2026 review of DMF-free routes).
What is a realistic solvent reduction to expect from wash reduction?
The most conservative bounded figure is over 50% total system solvent reduction, with crude purity falling from 93.2% को 91.4% when one post-deprotection wash and four needle washes are used (a vendor application note on the ACP65-74 10mer). Higher figures exist, up to approximately 80% on a 25 mmol liraglutide comparison, but they come from different scales and molecules and are not transferable (the wash-free SPPS work).
Should I ask for PMI or E-factor from a supplier?
दुबैको लागि सोध्नुहोस्, defined. PMI measures mass input per mass of product and averages 13,063 kg/kg for commercial and Phase 3 SPPS processes; E-factor measures waste per mass of product and is the correct basis for a waste-reduction claim (the ACS GCI Pharmaceutical Roundtable’s PMI analysis of 40 peptide processes). The two are not interchangeable, and a figure without its process stage, sequence length and scale cannot be compared.
निष्कर्ष
The pressure on peptide synthesis is real but narrower than the ETERNAL headline suggests. The binding constraint is Annex VII of the Industrial Emissions Directive, which already requires a solvent management plan from pharmaceutical manufacturing above 50 tonnes per year of solvent consumption, and the commercial constraint is Scope 3 reporting, where 95% of EFPIA’s surveyed member companies engage with suppliers on climate action (EFPIA’s survey of 23 member companies).
The cited multi-process study assigns approximately half of process mass intensity to downstream operations. This is not a universal share for every peptide, nor proof that chromatography always dominates. Evaluate the actual synthesis, purification and isolation balance when selecting an improvement.
The next step is a documentation request, not a specification change. Ask for a batch-level solvent mass balance against the Annex VII Part 7 categories, the analytical package for the incumbent route, and the revalidation scope if a solvent or wash change is proposed. A supplier that can answer those three requests is a supplier whose greener route you can actually qualify.
