Peptide Documentation Governance: An Audit-Ready Playbook for Supplier, Change, Impurity, Data and Release Controls

What did the FDA actually do about the six peptides, and is it final?

Peptide Pegylation Wholesale Peptides Research Nothing is final. The Federal Register notice opening the docket lists seven peptide categories for the July 23-24, 2026 meeting, each as free base and acetate: BPC-157, KPV, TB-500, MOTS-c, Emideltide (DSIP), Semax and Epitalon (Federal Register notice, Docket FDA-2025-N-6895, 2026-04-16). The same notice records 2,643 public comments filed on the docket before the meeting.

The vote tallies are reported, not published by FDA. Bass, Berry & Sims recorded six recommendations: BPC-157, KPV and TB-500 at 8-6, Semax at 8-5, MOTS-c at 7-5, Epitalon at 7-4, with Emideltide not recommended at 6-7 (the vote-by-vote record, 2026-07-31). McDermott Will & Emery independently reports the same tallies (a second read of the tallies, 2026). Two law-firm reports agreeing is corroboration, not an FDA transcript.

What follows a favorable vote is rulemaking. Section 503A permits a bulk drug substance only if it complies with an applicable USP/NF monograph, is a component of an FDA-approved drug, or appears on the 503A bulks list (the 503A eligibility tests themselves, page current 2026-03-26). Adding a substance to that list takes notice-and-comment rulemaking and a final rule, and Frier Levitt puts the timeline at 12 to 18 months or longer after a recommendation (the timeline to a final rule, 2026-10-14). An “Interim Access Policy for Compounding Using Certain Peptides” went to OMB on 2026-10-14; it is unpublished, not operative, and could be narrowed or withdrawn without rulemaking (what the still-unpublished policy could change, 2026-10-16).

What are FDA’s stated concerns about unapproved GLP-1 products?

The agency’s objection is chemical, h. FDA states that semaglutide sodium and semaglutide acetate are different active ingredients from the approved drug, that it has no information they share its chemical or pharmacologic properties, and that there is “no lawful basis for their use in compounding” (FDA’s stated concerns about compounded GLP-1s, 2026-01-10). The same page reports multiple adverse-event reports, some requiring hospitalization, tied to dosing errors with compounded injectable semaglutide, including cases where health-care professionals miscalculated doses.

As of 2026-05-31, FDA had received 990 adverse-event reports for compounded semaglutide and more than 730 for compounded tirzepatide. FDA notes that state-licensed pharmacies are not required to report, so the counts understate actual events; read them as a floor, not a rate.

The documentation implication is direct. If a salt form is a different active ingredient, then the counterion is a specification field, not a footnote. A record that says “semaglutide” without naming sodium, acetate or free base cannot be compared to the approved drug, cannot support a net peptide content calculation, and cannot be defended. The free-base/acetate pairing on the PCAC agenda makes the same field mandatory for every one of the seven substances discussed.

What must a peptide supplier qualification record contain?

A completed supplier questionnaire is not a qualification. ICH Q7 requires purchasing only from Quality-Unit-approved suppliers, against an agreed specification, with evaluation before approval that may draw on quality history, questionnaires, analytical comparison and, where justified, kev tshuaj xyuas; critical materials get more extensive evaluation and the manufacturer’s identity must be known (how Q7 supplier approval works in practice, APIC v17, 2024-11).

An on-site audit is a tool, not a mandate. APIC’s reading of Q7 is that audits are not required and their necessity should follow a documented risk assessment, with re-audit frequency set by supplier performance and criticality (how Q7 supplier approval works in practice, 2024-11). A file that contains an audit report but no risk assessment has the artifact without the reasoning.

The file itself has a field list. It should record the criticality assessment, a Quality-Unit-approved qualification plan, a fit-for-use assessment, the questionnaire and/or audit, specification and quality agreements, GMP/ISO certificates, the evaluation result approved by the Quality Unit before ordering, and the approved-supplier list, with a supplier risk score set immediately after qualification (what a supplier file has to hold, APIC v2, 2024-03).

The peptide layer sits on top. APIC’s guidance on full analysis is that it should cover every test in the user’s own specification, and that a supplier’s certificate of analysis may not align with the user’s specification; identity testing should use a discriminating method, and label inspection is not sufficient (how Q7 supplier approval works in practice, 2024-11). For a peptide that means running your own HPLC and mass spectrometry against your specification rather than filing the supplier’s CoA as the identity record.

How should change control work for a peptide sequence, supplier or scale change?

A supplier swap is a change to the material’s provenance, not a purchasing event. ICH Q10 requires a documented, risk-based pharmaceutical quality system across the lifecycle, with change management, a CAPA system whose investigation depth is commensurate with risk under Q9, management review, and control of outsourced activities and purchased materials, with the company remaining ultimately responsible (ICH Q10’s change-management and CAPA expectations, Step 4, 2008). APIC’s Q7 guidance adds that a change of source of a starting material or critical raw material should be handled under the change control procedure (how Q7 supplier approval works in practice, 2024-11).

Q6A may inform low-molecular-weight synthetic-peptide specifications but is insufficient for all peptide attributes or sizes. Q6B covers defined biological products and excludes chemically synthesized peptides. Use the actual product class and applicable peptide guidance.

The record fields a peptide change must carry: sequence, N- and C-terminus, h, daim ntawv tiv thaiv, lot, method ID, column, gradient, wavelength and reference standard. A counterion change from Stapled Peptides Factory trifluoroacetate to acetate alters the mass balance and the net content calculation, so it needs comparability data, not a note. A scale change alters impurity clearance, so it needs impurity profiles at the new scale.

E. A sequence edit made in a synthesis order without a version trail leaves the release specification pointing at a molecule the batch was not made to. Version control on the sequence is the change record.

How is an impurity or OOS result investigated and closed?

An out-of-specification result is a defined trigger, not a judgment call. FDA’s OOS guidance, a May 2022 Level 2 revision, covers all results outside specification or acceptance criteria in applications, DMFs, compendia or manufacturer standards, plus in-process laboratory tests outside specification (FDA’s OOS investigation guidance, page current 2024-08-01). An in-process test that fails is in scope.

An impurity result is out of specification only when it exceeds an applicable criterion. A new or trending peak can still warrant investigation, especially if identity or co-elution is unresolved. Investigate affected and potentially affected lots and assess disposition or recall from risk; neither 0.4% nor detection after shipment automatically determines that outcome. Peptide Order Factory

FDA has already put peptide impurities on the record. Its safety-risk listing cites “complexities with regard to peptide-related impurities and active pharmaceutical ingredient (API) characterization” as a concern for BPC-157, alongside possible immunogenicity for certain routes (FDA’s own safety-risk listing, page current 2026-04-22). The same page notes no human exposure data identified for KPV by any route. An impurity investigation file that cannot characterize the impurity is answering a question the agency has already asked.

What does data integrity require at the record level?

Audit trails are a feature; data integrity is a set of fields. 21 CFR 11.10 requires validated systems, the ability to generate accurate and complete copies, record protection, access limitation, secure computer-generated time-stamped audit trails retained at least as long as the records, operational and authority checks, and device checks; 11.50 sets what a signature must manifest, thiab 11.300 governs identification codes (Part 11’s control requirements, current eCFR text, tau txais rov qab 2026-10-15).

MHRA’s 2018 guidance supplies the organizing principles, including ALCOA+ and the requirement for true copies (MHRA’s data-integrity principles, Revision 1, 2018). The practical translation is that each record must be attributable, nyeem tau yooj yim, tam sim no, qub thiab raug, and complete, sib haum xeeb, ruaj khov thiab muaj.

Record-reconstruction test

  • Lot number resolves to the batch record and the release decision

  • Chromatogram resolves to the lot, the method ID and the column

  • Method resolves to its validation report and predefined criteria

  • Reference standard resolves to its certificate and expiry

  • Stability protocol resolves to the batch and the timepoint

  • Release decision resolves to a named signer, date and meaning

The failure mode is a certificate of analysis whose chromatogram cannot be matched to the lot it claims to describe. That is not a formatting problem. Under 11.10(y) the record must be reproducible in human-readable and electronic form for inspection, and a chromatogram with no lot linkage fails that test before anyone asks about the audit trail.

How are release specifications separated into purity, tus kheej, content, stability and assay?

The five attributes answer five different questions, and substituting one for another is the most common specification error in peptide work. Identity asks whether the molecule is the intended sequence; purity asks what fraction of the chromatographic area is the main peak; content asks how much peptide by mass is present; stability asks whether those hold over time; assay asks how much active substance is there against a reference.

Method validation is what makes each answer defensible. ICH Q2(R2) requires predefined performance criteria and a validation protocol stating intended purpose, measured attribute, performance characteristics and experimental design, with characteristics including specificity/selectivity, x, o, precision, detection and quantitation limits, and robustness (ICH Q2(R2)’s validation characteristics, 2023-11-30, corrected 2025).

Stability has a status trap. A consolidated ICH Q1 guideline replacing Q1A(R2) through Q1F and Q5C was endorsed at Step 2b on 2025-04-11 and issued by FDA as draft guidance in June 2025, but it is not for implementation, so Q1A(R2) (2003) remains the operative reference (the draft that will replace Q1A(R2), 2025-06). Under the draft, a new synthetic chemical entity still needs about three primary batches with 12 months long-term and 6 months accelerated data at submission, and the 25°C/60% RH long-term and 40°C/75% RH accelerated conditions are unchanged (the draft that will replace Q1A(R2), 2025-04-11).

Where K/M applies to a parenteral product, use the route-specific pyrogenic threshold and maximum dose over the specified interval. The common non-intrathecal parenteral value must not be extended to every oral, topical or research use. Select sterility tests according to product requirements.

How do you connect the six controls into one release decision?

The release decision is where peptide documentation governance either holds or fails, because it is the only point where all six controls are tested at once. A defensible disposition is a signed statement that traces to a supplier file, a change record, an investigation history, an intact data trail, a validated specification set and a stability protocol.

Run the gate in order and stop at the first failure:

  1. Supplier file complete, with the user’s own full analysis against the user’s specification, not the supplier’s CoA.

  2. Change record closed, with sequence version, counterion form and scale comparability documented.

  3. No open OOS or impurity investigation touching the lot, and no unresolved trend across prior lots.

  4. Data trail intact: chromatogram to lot, method to validation report, signature to 11.50 manifestation.

  5. Specification set separated into identity, purity, content, stability and assay, each with predefined criteria.

  6. Stability protocol current under the operative guideline, with the draft status recorded rather than assumed.

A release decision that rests on a certificate of analysis alone fails at step 1 and step 4 simultaneously. The certificate is a supplier’s claim; the record chain is your evidence.

Xaus lus

The regulatory picture as of October 2026 is unresolved by design. Six peptides carry non-binding recommendations, one did not, no substance has been added to the 503A bulks list, and an interim policy sits at OMB unpublished (FDA PCAC meeting page, 2026; what the still-unpublished policy could change, 2026-10-16). Enforcement did not wait for the rulemaking: five vendors received letters on 2026-08-24 (the five letters issued together, 2026).

The earned takeaway is a decision rule. Release a peptide lot only when a reviewer who was not in the lab can walk from the lot number to the chromatogram to the method to the reference standard to the stability protocol to a signed disposition, and can see at each step which governing text the record answers to. If any link requires your explanation, the record is not finished.

The next step is a gap assessment against the six-row matrix, run on the last three lots you released. Where a row has no document, that is the work. Where the document exists but does not name its governing text, that is also the work.
