Myristoyl Hexapeptide-16 is a synthetic lipopeptide. Synthetically, it is formed by covalently conjugating myristoyl to the N-terminus of hexapeptide-16.

Originating from a naturally secreted alkaline keratin-associated peptide within hair follicles, the natural peptide itself has short skin retention time and poor membrane penetration – a limitation addressed through lipid modification.

The introduction of the myristoyl group provides a hydrophobic tail capable of inserting into both cell membranes and the stratum corneum; significantly enhanced membrane permeability is the direct result.

Wide usage in various eyelash serums, anti-hair loss products and post-surgical repair creams demonstrates its utility. Its profile as a safer keratin-regulating agent (prostaglandin analogues being the primary comparison) is one of its primary advantages.

1.Primary Biological Activities:

Intracellular delivery is facilitated by a fatty acid mediated endocytosis process; upregulation of KRT1, KRT5 and KRT14 gene expression promotes keratin synthesis specifically in hair follicle cells, prolonging the anagen phase.

The lys-lys sequence charge cluster within its structure competitively binds TGF-βR II, inhibiting the transition signals towards catagen.

Simultaneous improvements in length and density of eyelashes and brows are the documented outcomes.

AQP-3 and filaggrin transcription are activated, strengthening the stratum corneum hydration barrier (reducing transepidermal water loss).

Production of various inflammatory mediators is modulated.

2.Keratin Gene Transcription

The basic peptide segment mimics KAP5/8 families, binding to NF-κB-like sites in the promoter regions of keratin genes.

This rapidly increased keratin synthesis enhances the hair shaft integrity and strength.

3.Extended Anagen Phase

The peptide effectively slows follicular regression, maintaining the anagen state.

Enhancing both length and density of eyelashes and eyebrows is observed, reduced hair shedding being another key effect; longer lasting hair growth is supported.

4.Strengthening the Stratum Corneum Barrier

Myristoyl Hexapeptide-16 binds to GPR40/FFA1 receptors on keratinocyte surfaces, activating the cAMP/PKA pathway, CREB (cAMP Response Element-Binding Protein) specifically being the downstream target; AQP-3 and filaggrin transcription follows as a result.

Increased natural moisturizing factor content is a direct outcome, better stratum corneum hydration function being achieved, a more effective skin barrier formed – transepidermal water loss is subsequently reduced.

Improved skin moisturization, enhanced barrier integrity and the alleviation of dryness and roughness are observed; damaged skin repair is within the scope of what this peptide can achieve.

5.Anti-inflammatory and Antioxidant Effects

Inhibiting various inflammatory signaling pathways (NF-κB being a key pathway) is one of its established effects, and pro-inflammatory cytokines (IL-6, TNF-α among others) are down regulated.

Skin redness, irritation and overall sensitive skin symptoms are mitigated by Myristoyl Hexapeptide-16.
