The core biological function of Acetyl Tetrapeptide-11 centers around maintaining and in some cases repairing, the structural and functional integrity of the Dermal-Epidermal Junction (DEJ). Two primary pathways are responsible for this mechanism of action:

Stimulating keratinocyte proliferation: Keratinocytes make up the epidermis themselves. Normal proliferation and subsequent differentiation are all aspects of a properly functioning skin barrier, structural stability, and to a large extent self repair. As a signaling peptide it directly promotes epidermal keratinocyte growth, increased cell density being one measurable outcome, accelerated regeneration being the end result of this process.

Promoting Glycoprotein Synthesis – specifically syndecan-1 and to a point type XVII collagen. Syndecan-1 (transmembrane heparan sulfate proteoglycan if I recall correctly) binds ligands (cell adhesion molecules, various growth factors) through its side chains; intercellular adhesion and all associated microenvironmental interactions are regulated. Type XVII collagen is a near essential component of hemidesmosomes. Reduced synthesis of either, with aging, leads to the dryness, laxity previously mentioned. Improving barrier function and radiance are both second order benefits of Acetyl Tetrapeptide-11.

Further scientific reports have shown a near complete blocking of neuromuscular transmission signals (by inhibiting SNARE receptors), reducing excessive release of both catecholamine and acetylcholine. Dynamic wrinkles are lessened by this action as well.

Clinical studies confirm that formulations containing acetyl tetrapeptide-11 (e.g., at a 3% concentration) significantly increase both skin thickness and firmness. One specific efficacy trial involved 19 female volunteers; the product in question was the sole active ingredient. Results showed a clear, measurable difference from a placebo control group – subjects using the acetyl tetrapeptide containing formulation had thicker, more firm skin.

This study robustly supports the ‘translation’ of various in vitro observed biological activities into tangible skin improvement. Evidence-based anti-aging is what this ingredient has established itself as being.
