Hexapeptide-33 (sequence: Ser-Phe-Lys-Leu-Arg-Tyr-NH₂, SFKLRY; W3 peptide) is a hexa-amino acid peptide.

First created by Novacell Technology through peptide library technology, This was made for cosmetic use and has several skincare benefits: the ability to induce angiogenesis; enhance skin elasticity; reduce wrinkles; and improve barrier function.

This peptide exerts its effects through binding to G-protein coupled receptors (GPCRs) widely expressed in human cells, particularly in skin keratinocytes where it participates in epidermal differentiation processes.

It is precisely because of the safety and efficacy of Hexapeptide-33, it has become a hot ingredient in many top anti-aging/brightening/repairing skin care products.

Hexapeptide-33 Related Research

The biological activities of Hexapeptide-33 are mostly mediated via the following mechanisms:

(1).Pro-Angiogenic

Through enhanced stimulation of vascular endothelial cells, Hexapeptide-33 accelerates microvascular density for superior skin blood flow and nutrient transport.

This is beneficial for dry skin, as it brightens dull complextion and speeds up tissue repair.

It is also thought to rev up the skin’s metabolic vigor.

(2).Promoting Collagen Synthesis

Hexapeptide-33 can activate dermal fibroblasts and increase collagen and elastic fiber synthesis, thus increasing skin compactness.

Some hexapeptide fractions have even been shown to increase the collagen gene expression and decrease the excess activation of MMPs which contributes toward delaying dermal ageing.

Aromatic sequence residues, such as phenylalanine and tyrosine were also found similarly to display antioxidation activity with free radical scavenging capacity, ultimately reducing photoaging effects by quenching free radicals due to photooxidative stress.

(3).Melanin synthesis inhibited

Hexapeptide-33’s brightening potential is believed to come from its ability to interfere with tyrosinase activity, or control melanocyte signaling.

Tyrosinase oxidizes tyrosine to dopa, then to dopaquinone and so on to eumelanin.

Most of the whitening peptides have backbone structure similar to natural tyrosinase inhibitory peptides to inactivate enzyme and depigment.

The tyrosine moiety of Hexapeptide-33 and its basic analogous capabilities with them can effectively compete with tyrosinase or related receptors (such as MC1R), leading to a reduction in melanin synthesis.

(4).Enhanced Wound Healing and Cell Migration

Positively charged amino acid residues such as Phe-Lys-Arg enhance peptide binding to cell membrane glycosaminoglycans (e.g., chondroitin sulfate), thereby prolonging local retention.

This increases the activity of enzymes, such as superoxide dismutase (SOD), which is an antioxidant and decreases the release of oxidative stress and inflammatory factors induced by UV radiation or other exogenous factors.

Additionally, it promotes keratinocyte migration and epidermal differentiation to enhance skin barrier repair, which is ideal for repairing damaged or sensitive skin.

(5).Conclusion

Hexapeptide-33 provides total skincare benefits in a triple-mode Whitening, Wrinkle-care and Wound-healing system.

Hexapeptide-33 uses: Hexapeptide-33 is a versatile bioactive peptide that can be designed into many anti-aging serums/creams and repair emulsions/masks, brighten/spot-fade products, premium eye care formulations.

And it is just so versatile, it can stimulate collagen production, repair the skin barrier, block melanin production and tackle periorbital concerns.
