Hexapeptide-9 (Hexapeptide-9, trade name Collaxyl™) is a synthetic bioactive peptide made up of six specific amino acids. The exact structure of this peptide is found within human collagen IV and XVII; these being closely associated with the key components of the skin’s basement membrane. Biotechnologically synthesized to mimic some, if not all, functions of natural collagen, purity and stability issues inherent in more ‘traditional’ natural extracts are eliminated. High stability and relatively easy formulation have led to its near universal adoption in various anti-aging skincare products.

Research Of Hexapeptide-9

Multiple multifaceted mechanisms of action are documented for Hexapeptide-9. A primary form of biological messaging is employed – activating and subsequently regulating the skin’s intrinsic synthesis and repair pathways.

Promoting Collagen Synthesis: Direct stimulation of dermal fibroblasts is one such mechanism; increased production of both Type I and Type IV collagen being the end result. Type I serves as the near constant structural support found in the dermis, resilience being a direct byproduct. Type IV (the ‘baby collagen’ if you will) links elasticity and more general smoothness to the skin.

Simultaneous elevation of both these collagens leads to effective remodeling of the underlying dermal architecture, lessening wrinkle depth and all round improvements in firmness.

Strengthening the Dermal-Epidermal Junction(DEJ): This is where Hexapeptide-9 truly sets itself apart. The DEJ (again, a physical bridge) is at least partially restored by it. Laminin-5 and Type IV collagen synthesis within this area is what makes Hexapeptide-9 so valuable to the end user.

Nutrient transfer, structural integrity and all that follows, is a constant fight against time and sagging.

Enhancing Skin Barrier and Hydration: Beyond its dermal actions, Hexapeptide-9 accelerates epidermal repair processes. Restoring damaged skin (post-acne marks, various forms of scratches, or even some post-aesthetic procedure effects) is one of its more valuable applications.

An in vitro efficacy study specifically showed the upregulation of the filaggrin (FLG) gene; epidermal differentiation and subsequently barrier function depend heavily on this protein. Filaggrin itself is a precursor to what are known as Natural Moisturizing Factors – increased presence being a direct indicator of both short and long term skin hydration, a near perfect end point of barrier defense.

Clinical studies have demonstrated increases in overall collagen production (up to 117% if I recall correctly) and more subtle but measurable improvements in elasticity.

Furthermore, research is now pointing towards cyclic hexapeptide-9 (CHP-9) as a potentially superior anti-aging solution. Traditional linear peptides have their place, but the cyclic structure offers greater stability, enhanced bioavailability and all round physiological activity.

Topical application of some form of this cyclic peptide is a very promising line of thought to either prevent or at least slow down skin aging. Confirming all of the above mentioned benefits through proper controlled studies is the next logical step.
