Research Of Oligopeptide-54

Oligopeptide-54 is a bioactive peptide prepared through solid-phase synthesis technology. The molecular structure has been engineered to at least partially mimic natural peptides’ functions.

As a core ingredient within many hair care products, combinations with other peptides (Decapeptide-10 being a prime example) and/or plant extracts are common. Liposome encapsulation is one technique used to improve both skin permeability and overall stability.

The primary biological activity centered around this peptide itself is hair care – stimulation of the follicle, all related forms of improved hair growth, and near complete resolution of certain pre-existing hair loss issues.

Enhancing melanin production within the follicle is also a reported effect (preventing graying being one of the more desirable end results).

Specific mechanisms of action are still being described by the suppliers in commercial literature, but pathways involving hair follicle cells are at least partially what Oligopeptide-54 is acting upon.

Oligopeptide-54 exerts these effects through multi-target regulation of the hair follicle microenvironment:

Inhibition of hair loss signaling – blocking the binding of BMP to its receptor (Bone morphogenetic protein receptors/BMPR) is the most well documented aspect.

Relieving the constant inhibitory effect BMP has on stem and dermal papilla cells is its key function.

Referencing the trade name again, ‘Nokkin’ seems to be a near perfect mimic of endogenous Noggin protein (directly binding BMPs to prevent activation).

Activating the Wnt/β-catenin pathway is a complementary, and to date still partially understood, positive regulatory effect of Oligopeptide-54.

Hair development and cycling are both positively regulated by Wnt; Overall, Oligopeptide-54 is a versatile peptide.

Promoting Hair Follicle Regeneration: Enhances scalp capillary microcirculation to nourish the hair follicles; stimulates matrix cell proliferation, directly leading to a strengthening of the hair shaft structure.

Antioxidant and Anti-inflammatory Effects: Reduces oxidative stress damage – scavenging various reactive oxygen species (ROS) being the primary mechanism.

Indirect regulation of the scalp microbiome balance is also observed, maintaining a more ‘healthy’ follicular environment.

Multiple suppliers market Oligopeptide-54 as part of their “hair growth peptide” series formulations; combinations with other bioactive peptides (Oligopeptide-2, Decapeptide-18 if I recall correctly) are commonly used for synergistic effects.

The hair growth cycle itself is a long term process (2-5 years being typical), age, gender and to some extent season all play into it.

Long term use is what will ultimately show positive, measurable effects from Oligopeptide-54.
