Oligopeptide-2 is a bioactive peptide with high interest for the cosmetics sector, primarily used in anti-wrinkle and anti-aging products.

It is based on a fragment from the C-terminal region of the collagen α1 chain, involved in collagen–laminin–integrin crosslinking.

The endogenous peptide fragment, however, is quickly broken down by proteases on the skin surface, hindering its effective penetration through the stratum corneum.

This has spawned a number of derivatives, the most successful of which is Oligopeptide-2.

A palmitoyl moiety is added to increase the lipophilicity of the peptide, facilitating its penetration through epidermal lipid bilayers and thus increasing overall stability for inclusion in cosmetic formulations.

Consequently, Oligopeptide-2 has become a vital component in top quality cosmetics.

1.Mechanism of Action and Benefits

Oligopeptide-2 induces fibroblast maturation, activates downstream intracellular signaling pathways, remarkably increases the de novo synthesis of type I and III collagens, efficiently stimulates fibronectin and glycosaminoglycan production and exhibits chemotactic activity, promoting fibroblast migration and aggregation at target sites, thereby accelerating skin recovery.

With continued use, Oligopeptide-2 reduces the depth and density of facial wrinkles, improves skin texture and strengthens the skin barrier function.

2.Promotes Collagen Synthesis

Oligopeptide-2 binds to specific receptors on fibroblast membranes, activating the TGF-β signaling pathway.

This leads to the activation of collagen synthesis-associated mRNA, which promotes highly effective and rapid intracellular production of precursor molecules for type I and III collagen.

These are further processed into complete and stable networks of collagen fibers.

This increases collagen fiber density and quality, resulting in improved skin elasticity and firmness and reduced wrinkles.

3.Stimulates Production of Fibronectin and Glycosaminoglycans, Enhancing Skin Hydration

In addition to collagen synthesis, Oligopeptide-2 stimulates the production and activation of fibronectin and glycosaminoglycans.

It enhances fibronectin synthesis and hyaluronic acid synthase activity, resulting in a gel matrix that improves the skin’s moisture retention capacity.

As a result the skin appears plumper, fuller and more hydrated.

4.Chemotactic Effect

Oligopeptide-2 has strong chemotactic properties, guiding fibroblasts and their precursor cells to migrate directionally toward damaged skin areas.
