Un valore di purezza non è una proprietà del materiale. Problema 1: il numero è definito dal metodo. Problema 2: l’identità di massa non è purezza. La LC-MS conferma l'accurata massa molecolare abbinata alla separazione cromatografica, e MALDI-TOF fornisce una rapida impronta digitale della massa intatta. I fallimenti documentati sono identità, errori di etichettatura e contaminanti, carenze di purezza non marginali. In 2026, Victoria’s Chief Health Officer linked six cases of acute liver toxicity to unapproved peptide products labelled Retatrutide, noting the clinical presentation suggested “additional contaminants may be contributing” (RACGP newsGP, 22 Giugno 2026). By late August, seven Victorian patients had been hospitalised for acute liver toxicity after using a counterfeit of retatrutide, and Canberra Health Services reported liver damage without releasing a count (ABC News, 24 agosto 2026). In both reports the contaminant cause is stated as a hypothesis, not a confirmed finding.

Those figures reached this article through a secondary summary whose own text says they should be confirmed against the primary guideline, and the primary PDF could not be read here, so treat them as a working reference and check the source document directly before you write them into a specification.

Impurity control is anchored to the Ph. Euro. general monograph “Substances for Pharmaceutical Use”, and the guideline requires it to be scientifically justified by batch data rather than aligned to a theoretical threshold. That distinction matters when you read a certificate of analysis: the categories are specified identified, specificato non identificato, unspecified and total, and a single “purity” figure collapses all four.

Non-peptide contaminants sit outside that table and need their own tests. Endotoxin is measured by USP <85> / LAL, sterility by USP <71>, residual solvents by headspace GC against ICH Q3C, the TFA counterion by ion chromatography, CE or ¹⁹F NMR, counterion identity by ion chromatography, and moisture by Karl Fischer titration. Commercial specifications are published, for example by GenScript, with endotoxin at ≤0.01 EU/µg and TFA held below 10%, down to below 1% for some grades.

One more number belongs in the same conversation: net peptide content is not purity. Low NPC is expected for basic and hydrophilic peptides even at high purity, and Bachem’s QC guide notes the batch-to-batch variability this creates, so both figures have to be considered when you prepare a solution.

Peptide supply chain traceability is the criterion that makes every other test auditable, and it is the one buyers skip most often. A purity figure, an identity result and an impurity profile are only as good as the record that ties them to a specific vial.

  1. Request the peptide certificate of analysis with method parameters. Ask for the chromatogram, the peak table, and the column, gradient and detection wavelength behind them. Quick win: same day.

  2. Check identity evidence. Confirm the package reports observed versus theoretical mass and states sequence coverage. Quick win.

  3. Check impurity reporting. Ask which thresholds the supplier applies for reporting, identifying and qualifying impurities, and whether batch-to-batch variability is described. Longer-term.

  4. Confirm the non-peptide panel. Endotossina, sterilità, solventi residui, counterion and moisture should be reported where your intended use requires them. A complete package for one lot typically contains the CoA with method parameters, a representative chromatogram and peak table, identity data, impurity results against stated thresholds, the applicable non-peptide panel, and a lot number that matches the label and your receiving record. Measure two things: the proportion of lots arriving with a complete package, and the rate of discrepancies between the label and the CoA. Expect the first supplier comparisons to be decisive within one or two procurement cycles.

Two things this research does not establish deserve stating plainly. No source here gives a verified purity percentage measured on a retatrutide-labelled retail sample, and no source gives a verified rate of end-user certificate-of-analysis forgery. The prevalence of the problem is unproven.

The conventional view also still holds in places. For early-stage qualitative work such as Western blotting, a lower-grade material with a documented method may be entirely adequate, and Bachem’s QC guide grade tiers remain a reasonable starting heuristic when the intended use is qualitative.

NO, because a grade tier and an identity test answer different questions. A purity grade tells you how much of the material in the vial is the stated compound; it does not tell you that the stated compound is the one in the vial. Use both: a grade tier as a starting filter, identity confirmation as the test that actually closes the gap.

That request is the whole transition, and the response is the finding. A supplier that cannot produce method parameters and a chromatogram for a past lot has told you something about how that lot was qualified, and about what the documentation will look like for the next one.

If that standard becomes routine, a peptide certificate of analysis stops being a marketing artifact and becomes a checkable record: a document a reviewer can audit, not a number a buyer has to trust. That is the future state worth working toward, and it does not require new regulation to start. It requires buyers to ask better questions before they order.
